Gıda ve yem ürünlerinde pestisit kontrolünün izlenmeye başlanmasıyla birlikte insan sağlığı
ile ilgili endişeler giderek artmıştır. Veteriner tıbbında ve aynı zamanda hayvan yemi
üretiminde yaygın olarak kullanıldıkları göz önüne alındığında, gıda güvenliği konusundaki
endişelerin artması haklıdır. Bu çalışmada, kanatlı eti örneklerinden farklı pestisitlerin
(dimetoat, imazalil, metil parathion, metalaxyl, methiocarb ve fludioxonil) ekstraksiyonu ve
analizi için QuEChERS katı faz ekstraksiyonu ve dağıtıcı temizlemenin bir kombinasyonu ve
ardından gaz kromatografisi rapor edilmiştir. –kütle spektrometrisi tespiti. Analiz için tartılan
numunelerin kütlesi, santrifüj öncesi girdaplama süresi, ekstraksiyon çözücü tipi ve tuz ilavesi
gibi ekstraksiyon verimini etkileyen bazı önemli parametreler incelenmiştir. Kanatlı eti
örnekleri, proteinleri ve yağları çökeltmek için optimize edilmiş asetonitril/su çözeltisi ile
işlendi. Asetonitril-su (80:20, v/v) ile altı pestisitten daha yüks...ek bir geri kazanım oranı elde
edildi. Bu rasyon, gerçek numuneler için optimal ekstraksiyon çözücüsü olarak seçilmiştir.
Analitlerin kromatografik analizi, elektron iyonizasyonlu Clarus SQ8T kütle spektrometresi
(PerkinElmer, Waltham, MA, ABD) ile bir gaz kromatograf sistemi Clarus 680 üzerinde
yapıldı. Spitless/split enjektör ile donatılmış GC/MS cihazı. Geliştirilen yöntem, optimize
edilmiş deneysel koşullar altında seçiciliği, duyarlılığı, doğruluğu, kesinliği ve doğrusallığı
açısından doğrulanmıştır. Optimum ekstraksiyon koşulları altında, sırasıyla 1.02 – 1.52 ve 3.4
– 5.1 μg kg-1 aralığında saptama ve niceleme limitleri elde edildi. Kalibrasyon eğrileri,
belirleme katsayıları ≥ 0.99 ile doğrusaldı. Kör tavuk eti numune solüsyonu ile hazırlanan
kalibrasyon eğrileri, matris uyumlu solüsyonlarla hazırlanan kalibrasyon eğrileri ile
karşılaştırılmıştır. Göreceli standart sapma, intra– (n = 5) ve günler arası (n = 5) kesinlik için
%15'in altındaydı. Son olarak 50 adet tavuk eti örneğinde pestisit kalıntısının belirlenmesi için
yöntem başarıyla uygulanmıştır. Test edilen tüm numuneler, dondurulmuş tavuk fileto
numuneleridir. Bu yöntemi kullanarak, tavuk fileto örneklerinin %6.0'ında 5.4 ila 8.9 μg kg-1
konsantrasyon aralığında pestisit imazalil belirledik. Imazalil, narenciye meyvelerinde hasat
sonrası kullanım ve arpa ve buğday tohumlarının ekim öncesi uygulamaları için sistemik bir
fungisittir. Ancak imazalil'in tavuk kuluçka tedavileri için gıda dışı bir kullanımı da vardır.
Tavuk etinde imazalil izlerinin bulunmasının sebeplerinden biri de bu olabilir.
Since pesticides control started to be monitor in the food and feed products concern for human
health has become more and more. Given that they are widely used in veterinary medicine,
but also in the production of animal feed, increasing concern for food safety is justified. In
this study, a combination of QuEChERS solid phase extraction and dispersive clean-up has
been reported for the extraction and analysis of different pesticides (dimethoate, imazalil,
methyl parathion, metalaxyl, methiocarb, and fludioxonil) from poultry meat samples
followed by gas chromatography–mass spectrometry detection. Some important parameters
affecting the extraction efficiency were studied such as the mass of the weighed samples for
analysis, the time of vortexing before centrifugation, the type of extraction solvent and the
addition of salt. The poultry meat samples were treated with optimized acetonitrile/water
solution to precipitate proteins and fats. A higher recovery rate of the six pesti...cides was
obtained with acetonitrile-water (80:20, v/v). This ration was chosen as the optimal extraction
solvent for real samples. Chromatographic analysis of the analytes was performed on a gas
chromatograph system Clarus 680 with Clarus SQ8T mass spectrometer with electron
ionization (PerkinElmer, Waltham, MA, USA). GC/MS instrument equipped with a
spitless/split injector. The developed method was validated in terms of its selectivity,
sensitivity, accuracy, precision and linearity under the optimized experimental conditions.
Under the optimum extraction conditions, limits of detection and quantification were obtained
in the ranges of 1.02 – 1.52 and 3.4 – 5.1 μg kg-1, respectively. The calibration curves were
linear with coefficients of determination ≥ 0.99. Calibration curves prepared with the blank
chicken meat sample solution were compared with calibration curves prepared with matrixmatched
solutions. The relative standard deviation was under 15% for intra– (n = 5) and
inter–day (n = 5) precisions. Finally, the method has been successfully applied for the
determination the pesticides residue in the 50 chicken meat samples. All tested samples were
frozen chicken fillet samples. Employing this method, we identified the pesticide imazalil in
6.0% of chicken fillet samples in the concentration range of 5.4 to 8.9 μg kg-1. Imazalil is a
systemic fungicide for post-harvest use on citrus fruits, and preplanting seed treatments of
barley and wheat. But imazalil also has a nonfood use is for chicken hatchery treatments. That
can be one of the reasons for the presence of traces of imazalil in chicken meat.
